The activities of several enzymes that have previously been utilized to identify intact mitochondria have been examined and critically evaluated in subcellular fractions obtained from rat cardiac homogenates.The localization of the enzymes investigated (succinate dehydrogenase, fumarase, isocitrate dehydrogenase (NADP- and NAD-dependent), NADH oxidase and monoamine oxidase) have been compared directly with the visual evidence obtained from electron microscopy studies. In addition the quantitative estimation of the mitochondrial histochemical stain Janus Green B was correlated with these studies.It was concluded that the most satisfactory mitochondrial indicator enzymes in rat heart are succinate dehydrogenase and isocitrate dehydrogenase (NAD-dependent).
