The fluoroquinolone susceptibility and molecular epidemiology of Mycobacterium tuberculosis strains isolated from 16 pulmonary tuberculosis patients at the University Teaching Hospital Chest Clinic, Lusaka, Zambia, were investigated [date not given]. The susceptibility of the isolates to isoniazid, rifampicin, ethambutol and streptomycin, and the minimum inhibitory concentrations to ciprofloxacin, sparfloxacin and levofloxacin were determined. The fluoroquinolone resistance-determining region (QRDR) in gyrA gene was sequenced and analysed. The results showed that 3 strains were resistant to isoniazid, rifampicin and/or streptomycin. All strains were susceptible to ciprofloxacin, levofloxacin and sparfloxacin. However, a unique gyrA gene variation was identified in the isolates. One strain had a mutation (T73A) in QRDR. In addition, a SerThr substitution at codon 88 was observed in 13 strains (81.25%), all of which were susceptible to fluoroquinolones. It is suggested that the gyrA variation observed in this study may provide useful epidemiological information for future studies on fluoroquinolone therapy of resistant M. tuberculosis infection.
