Interstitial cells from female rats aged 25-26 days were cultured in a serum-free medium. In the presence of LH or HCG, but not FSH, steroid synthesis was about 200 times greater than in their absence. Of the steroid secreted, 96% was androgen, and of this 98% was androsterone. The stimulation of androsterone secretion by LH or HCG was dose-dependent. The effect of LH on androsterone secretion was mimicked, in a dose-dependent manner, by PGE2, cholera toxin and 8-bromo cAMP. In in vivo experiments, administration of HCG induced a pattern of androgen response similar to than found in vitro.
