Materials and MethodsFor the present investigation more than 500 samples of duodenal tissues were examined. Most of these, however, were unsatisfactory: this was either because the mucosae showed pathological changes or because the sections were not cut or stained in a suitable manner. Fifty-six samples proved acceptable for the purpose of mitotic counts. These were obtained from 47 male and 9 female patients, ranging in age from 28 to 75 years, operated for various gastro-intestinal lesions. All these specimens were at least 4 mm from the sites involved in the pathological process. No significant change from the normal histology was observed in the mucosae of the tissues used; only a few showed evidence of subacute or chronic inflammation. Pieces of tissue, measuring approximately 0.5 x 1 cm. were fixed in Davidson modified fixative or in 10% formalin and blocked in paraffin. The sections were cut at 6 to 7 micra, and stained with hematoxylin and eosin. The method used to enumerate mitoses followed basically that devised by Leblond and Stevens [1948], Because of the uneven distribution of dividing cells, only areas where the villi were cut approximately throughout their whole length were used for the count. The cells were coifnted in consecutive fields covering all areas from the base of the crypts of Lieberkulin to the top of the villi. With a hand tally counter and a binocular microscope, under oil immersion, at least 2000 nuclei of resting and dividing cells were counted in each specimen. Paneth cells and argentaffine cells, which do not undergo cell renewal (Leblond and I talker [1956]). were excluded from the counts. The number of dividing cells was recorded as per centage of the total cell-count.
