The Properties of A Protease Activated by High Pressure from Sporosarcina sp. strain DSK25 Isolated from Deep-sea Sediment

From a deep-sea investigation in 1991 by members of the Japan Marine Science and Technology Center (JAMSTEC), deep-sea mud samples were obtained from the Japan Trench at 6500 m depth (40'06.8"N 144'11.0"E), using the manned submersible "SHINKAI 6500". The deep-sea bacterium, strain DSK25 was isolated from the mud samples, and several of its properties were determined. Strain DSK25 belongs to the genus Sporosarcina sp., and produced an extracellular protease. The protease from strain DSK25 had optimal activity at around pH9 and 40°C and its activity was inhibited completely by phenylmethylsulfonyl fluoride (PMSF). It is suggested that this protease is an alkaline serine protease. The activity of the protease from strain DSK25 was enhanced by elevated pressure, whereas other commercial proteases showed no pressure enhancement. This suggests that the protease may be adapted to the high pressure environment found in the deep-sea.
