Natural killer (NK) cells are large granular lymphocytes that spontaneously lyse certain tumour and virally infected cells [1, 21. Although NK cells share a number of morphologic, phenotypic and functional features with cytolytic T-lymphocytes, they do not productively rearrange the genes for the T-cell antigen receptor (TCR), and do not express a cell-surface TCR complex [3, 41. The demonstration that freshly isolated NK cells do not express a TCR clearly distinguished NK cells from T-lymphocytes and served to resolve a long-standing controversy that stemmed from the ability of cytolytic T-cells to acquire, upon activation, a cytolytic spectrum similar to NK cells. The full range of physiological functions of NK cells has not been determined, but probably is not limited to host defence against malignancies and viral infections [1, 21. Upon activation, NK cells produce an array of cytokines, suggesting that these cells have complex regulatory roles in addition to their cytolytic functions [5-71.The intracellular signals that trigger the cytolytic activity of NK cells, and the cell-surface receptors that transduce those signals during recognition of target cells, have not been defined. To approach these problems, we have used RNK-16, a rat leukaemia line with the phenotypic and functional characteristics of rat NK cells [8, 91. RNK-16 cells do not express a TCR and, like freshly isolated rat NK cells, lyse YAC-1 target cells [8, 91. An advantage of this model system is that RNK-16 cells are a clonal population free of the contaminating cells that can complicate studies of fresh NK cells.
