Role of phosphoinositides in transmembrane signaling.

In recent years, there has been widespread interest in the agonist-stimulated breakdown and turnover of myo-inositol-containing phospholipids. Although these lipids comprise< lo% of the total cellular phospholipid, their stimulated turnover accounts for a considerably higher percentage of total phospholipid turnover. The phosphoinositide field is currently the number one field in biochemistry in the number of citations (excluding molecular biology). On the basis of one abstract service, the number of papers published in this area is~ 150 papers per month or 5 papers per day. It now seems clear that the phosphoinositide effect is a multiregulatory mechanism involving the release of various moieties of the parent molecules, which serve as second messengers subserving myriad changes in the cell elicited by receptor activation by a wide variety of agonists. In addition to the well-established second messengers, inositol 1, 4, 5trisphosphate(1, 4, 5-IP3) and l, Z-diacylglycerol (DAG), evidence is mounting for additional second messengers. Numerous reviews on this subject are appearing monthly (eg, see Refs. 2, 39, 50, 53, 128, 185,304,676,796), although recently they have tended to focus on one aspect of the phosphoinositide response or one cell type. In this review, we tried to cover most aspects of the field. We hope that the extensive bibliography presented here may be useful for the neophyte as well as for the more experienced investigator.
