Comparative study of the biological characteristics of two human ovarian carcinoma models in nude mice

Objective:To establish intraperitoneal(IP) injection model and subcutaneous(SC) injection model of human ovarian carcinoma in nude mice and compare their biological characteristics.Methods:The cell suspension derived from A2780/DDP were injected intraperitoneally or subcutaneously in nude mice to establish human epithelial ovarian carcinoma models,and their growth behavior and biological characteristics were observed.Distribution of cell cycles were analyzed by flow cytometry(FCM).Expression of LHRH-R were investigated by immunohistochemistry.Results:The human ovarian carcinoma models were established successfully.The success rate of transplantation in both groups were 100%.The median survival time lengths in IP group and SC group were 30.000±1.095 days and 41.000±0.894 days,respectively.FCM analysis showed the distribution of cell cycles of xenografted tumors in two groups were the same as those of A2780/DDP cells.Histology and electron microscopy showed the same characteristics and morphological properties in the two groups,and immunohistochemistry staining of LHRH-R were both positive.Conclusion:Human ovarian carcinoma models in nude mice are easy to establish.And the IP model may be an ideal model for treatment research of ovarian carcinoma as it simulates the intraperitoneally dessiminating behavior of human ovarian carcinoma in the patients.
