Two forms of a calcium activated neutral protease (CANP) were purified from rabbit skeletal muscle to homogeneity. One is a low calcium-requiring form (CANP I) and the other is a high calcium-requiring form (CANP II). Previously we reported that CANP II was converted to the low calcium-requiring form (,uCANP) by autolysis. We compared ,uCANP with CANP I to determine their identity. They were similar with respect to sensitivity to calcium ions, reaction with protease inhibitors and pH activity profiles, but their molecular weights and stabilities were different. We conclude that CANP I and ,uCANP are different forms of a protease which is active at low concentrations of calcium ions.
