目的:本研究旨在探讨天疱疮患者血清中白细胞介素1α(IL-1 α)、趋化因子单核细胞趋化蛋白1(也称为趋化因子[C-C基序]配体2 [CCL 2])和C-X-C基序趋化因子配体2(CXCL 2)水平的可能变化。研究方法:采用酶联免疫吸附法(ELISA)检测57例PV天疱疮患者(其中进展期42例,缓解期15例)和31例健康对照者血清中IL-1 α、CCL 2和CXCL 2的表达水平。采用独立样本t检验对两组进行比较。采用单因素方差分析进行多组比较,采用事后最小显著性差异检验检测多组间差异。结果如下:进展期胃癌患者血清CCL 2和IL-1a表达水平均显著高于对照组(分别为[2.69 ± 0.23] ng/mL vs. [2.55 ± 0.28] ng/mL,P = 0.043; [0.62 ± 0.27] ng/mL vs. [0.48 ± 0.23] ng/mL,P = 0.038)。CXCL 2在进展期患者血清中的表达水平明显高于缓解期患者([61.70 ± 46.38] ng/mL vs. [24.97 ± 18.46] ng/mL,P = 0.037)。性别、疾病类型、疾病严重程度、治疗方式和黏膜受累对患者组和对照组血清中IL-1 α、CCL 2和CXCL 2的表达无显著影响(均P > 0.05)。结论:IL-1 α、CCL 2和CXCL 2与天疱疮的发生、发展密切相关,可能与疾病的活动性有关。
Objective: This study was performed to explore the possible changes of the serum levels of the cytokines including interleukin 1α (IL-1 α), chemokine monocyte chemotactic protein 1 (also known as chemokine [C-C motif] ligand 2 [CCL2]), and C-X-C motif chemokine ligand 2 (CXCL2) in patients with pemphigus. Methods: The expression levels of IL-1 α, CCL2, and CXCL2 in the serum of 57 patients with pemphigus PV (including 42 patients in progressive stage and 15 patients in remission stage) and 31 healthy controls were examined by enzyme-linked immunosorbent assay. The indepent-samples t-test was used to compare the two groups. One-way analysis of variance was used for multiple-group comparisons, and the post-hoc least significant difference test was used to detect differences among multiple groups. Results: The serum expression levels of CCL2 and IL-1a were all significantly higher in the patients in progressive stage than in the controls ([2.69 ± 0.23] ng/mL vs. [2.55 ± 0.28] ng/mL, P = 0.043; [0.62 ± 0.27] ng/mL vs. [0.48 ± 0.23] ng/mL, P = 0.038, respectively). In addition, the serum expression level of CXCL2 was significantly higher in patients in progressive stage than in in the remission stage ([61.70 ± 46.38] ng/mL vs. [24.97 ± 18.46] ng/mL, P = 0.037). Sex, disease classification, disease severity, treatment, and mucosal involvement had no significant influence on the expression of IL-1 α, CCL2, or CXCL2 in the serum of patients groups and controls (all P > 0.05). Conclusion: IL-1 α, CCL2, and CXCL2 are heavily involved in the occurrence and development of pemphigus and may be related to the activity of the disease.
