By the technique of autoradiography underin vitroconditions it has been possible to demonstrate that some of the red blood cells of the 5-day chick embryo take up3H-thymidine in the synthesis of DNA and essentially all of them take up3H-uridine as a RNA precursor.There is a rather great synthesis of DNA in certain early cells of the definitive erythrocyte line. This seems surprising, because these cells have not been found to undergo mitosis. A weak label was found in red blood cells from embryos as old as 14 days of incubation.RNA labeling was found to occur first in the nucleus, then spread into the cytoplasm with time in the blood cells of the 5-day embryo. No uptake of3H-uridine was observed in mature primitive erythrocytes, but was apparent in the early polychromatic cells of the definitive line.The significance of the observations in thesein vitroexperiments in relation to the total synthetic capacity of the erythrocytes of the chick embryo during development is discussed.
