The tachykinin peptides, substance P (SP) and neurokinin A (NU, also called substance K), are generated from a preprotachykinin (PPT) protein precursor, encoded by the PPT gene.'-'The primary PPT mRNA transcript is differentially processed into three mature PPT mRNAs: a, p, and y. I4 In the rat, the p-and y-PPT mRNAs, each encoding SP and NKA, are predominant: SP and NKA are present in rat anterior pituitary glandss2" Our previous work," recently confirmed by demonstrated that the PPT gene is expressed in rat anterior pituitary, suggesting that this tissue is a site of tachykinin synthesis. After puberty, male rat anterior pituitaries have more SP and NKA than do anterior pituitaries of female rat^.^.^.'"'^.'^ A comparable difference in adenohypophyseal PPT mRNAI7 suggests that sex differenccs in tachykinin concentrations arise from sex differences in PPT gene expression.
