Metabolism of arachidonic and linoleic acids by prostaglandin H synthases and lipoxygenases has been implicated in modulating cell proliferation, differentiation, and transformation. Increased production of these lipid compounds is associated with many tumor cell types and these products are described as regulating immune function, tumor growth, promotion, and metastasis. Inhibition of cyclooxygenase and lipoxygenase metabolic pathways is associated with a reduction in the incidence and mortality of some human forms of cancer. Our work has characterized linoleic and arachidonic metabolism as playing a central role in transducing the epidermal growth factor (EGF) mitogenic signal in several fibroblast cell lines.
