Cocaine (COC) and Its principal metabolite In man, benzoylecgonine (BE) were determined by quantitative gas-liquid chromatographic methods. Nitrogen detection was used for COC and electron capture detection for BE following extraction from human plasma. Using flame Ionization detection, COC and BE were simultaneously determined from human urine. The limits of detection for COC underivatlzed andBE as the pentafluorobenzyl bromide derivative In plasma were 10 and 5 ng/mL, respectively. In urine the sensitivity limits of the sllyl derivatives of COC andBE were 0.5 and 1.0 Mg/mL, respectively. The coefficient of variation ranged between 0. 9-2.2% and the coefficient of determination was 0.99 for these methods. Data on plasma and urine concentration of COC and BE collected over a time period of 24 h from three human subjects who received 1.0 to 1.9 mg/kg cocaine’HCI1. v. are presented.
