An Attempt to Achieve Efficient Nerve Regeneration After Rat Femoral Nerve Injury

In spite of the accurate neurorrehaphy with the application of microsurgical techniques, full functional recovery after peripheral nerve injury is still unlikely. The theory of selective nerve regeneration indicates that creating a small gap between severed nerve stumps by a nerve conduit can promote accurate axonal regeneration and functional recovery. However, lacking cellular and neurotropic support could diminish the growth promoting effect of conduits. In the current study, a 5 mm defect was created on the main trunk of rat femoral nerve. The defect was bridged using autologous nerve grafting, nerve conduit with basic fibroblastic growth factor and stem cells or nerve conduit with cell culture medium. 12 weeks after the surgery, the treatment effect was evaluated by the weight of right quadriceps femoris muscle, electrophysiological analysis, retrograde labeling as well as transmitting electronic microscopic tests. At the final analysis, no significant differences were found comparing the weight of right quadriceps femoris muscle, electrophysiological analysis and transmitting electronic microscope analysis between autologous nerve grafting group and nerve conduit bridging with stem cells and neurotropic factor group. Retrograde labeling revealed better selective axonal regeneration with conduit bridging than autologous nerve grafting. It can be concluded from the current research that, proper combination of nerve conduits, stem cells and fibroblast growth factor can effectively facilitate the accurate axonal regeneration and functional recovery after peripheral nerve injury.
