Short-term perfusion of the isolated dog gastrocnemius-plantaris muscle with H2O2 resulted in a decrease in steady-state muscle O2 consumption and isometric tension generation. H2O2 converted intracellular myoglobin to products incapable of combination with O2, but had no deleterious effect on neuromuscular transmission or on mitochondrial oxidative phosphorylation. Functional intracellular myoglobin is important in maintaining O2 consumption and tension generation in exercising skeletal muscle.
