In 2003, the first biologic therapy (omalizumab) for asthma was US Food and Drug Administration approved and immediately revolutionized the treatment of patients with severe asthma, including patients with evidence of type 2 (T2) inflammation. In the years since approval, additional monoclonal antibodies against T2-high asthma targets have widened the therapeutic landscape and include antagonists to IL-5 (mepolizumab and reslizumab), the IL-5 receptor (benralizumab), and the IL-4 receptor (dupilumab). These medications have shown considerable efficacy, yet some patients with severe asthma have no initial response, cannot tolerate the adverse reactions, or lose therapeutic response with time. Although there are many medications in development to improve asthma management, the following articles outline three new promising and not-so-promising approaches for the treatment of severe T2-high asthma.
