During the last 15 years, there has been a rapid advance in our ability to discriminate among individual proteins in complex mixtures, and the sensitivity of analytical protein detection methods has increased dramatically. This has led to the recognition of a large number of proteins that could not previously be detected and has opened up new possibilities for investigations of proteins. With such modern analytical techniques being used, the complexity of the composition of salivary proteins has become increasingly apparent and has facilitated studies on protein polymorphism and determination of patterns of inheritance. In parallel, the explosive advance in DNA recombinant technology has made it possible for the structure of the genes to be analyzed for a number of proteins, including those in saliva. While our understanding of the genetics of salivary proteins is far from complete, present knowledge gives us a much better insight into the manner in which the information encoded in the genes leads to the structure of the secreted proteins. Numerous studies have been done on human as well as animal salivary proteins, but this review will be limited to the genetics of proteins that, as far as is known, are synthesized in human salivary glands.
