Abstract The diuretic and natriuretic responses to acute oral salt loads were compared in two strains of rats with genetically determined opposite susceptibility to hypertension. Adult female rats of the Brookhaven R and S strains were given 6 ml. intragastric loads of 0.6M; 0.3M; and 0.15M NaCl solutions. Hypertonic saline loads elicited equivalent diuretic and natriuretic responses in both the hypertension-prone (S) and the hypertension-resistant (R) rats. The diuretic response to hypertonic saline was characterized by low but comparable urine osmolalities in both strains: S: 758 ± 34; R: 779 ± 34 mOsm.; (p > 0.1). Isotonic loads produced a significantly higher diuretic and natriuretic response in the S than in the R rats: V(milliliters per 2 hours): 2.3 ± 0.7 vs. 0.8 ± 0.4 (p 0.1). The data suggest an abnormal concentrating mechanism which is unmasked during isotonic volume expansion and was not observed during hypertonic saline diuresis. The occurrence of an increased sodium and water excretion in young normotensive, but hypertension-prone rats, suggests that the anomaly precedes the advent of elevated blood pressure.
