The association of human papillomaviruses (HPVs) with anogenital cancer is now supported by such overwhelming evidence that even the most sceptical critic of a virus/cancer hypothesis must take note. Nevertheless, when the data are examined objectively, it becomes clear that as with so many, perhaps all, cancer etiologies that have been studied, more than one factor is involved in the progression to malignancy. HPV may be the major player in this disease, may be necessary to initiate and maintain the process, but for the eventual manifestation of invasive carcinoma may not be sufficient. In vitro studies of the effects of HPV gene expression on primary human cells in culture have provided a basis for evaluation of the phenotypic changes resulting from interaction of the viral gene products with cellular genes.
