EFFECT OF CALCIUM ON INTRACELLULAR SODIUM + POTASSIUM CONCENTRATIONS IN PLANT + ANIMAL CELLS

The intracellular Na and K concentrations of mouse pancreas,R. pipiens(frog) oocytes, HeLa cells, ascites tumour cells, yeast andE. coliwere compared after incubation in Ca-containing medium and in Ca-free EDTA medium. With the exception ofE. coli, intracellular Na was always less than extracellular Na. All cell systems maintained a K gradient in the presence of extracellular Ca, the ratio of intracellular: extracellular K ranging from 6 to 80. In Ca-free EDTA, all cell systems gained Na and lost K. This was not prevented by the presence of Mg and was readily reversed by transfer of the cells to Ca-containing medium. These results suggest that extracellular Ca ion is required for the maintenance of low intracellular Na and high intracellular K in living systems. The few reported exceptions to this requirement are attributed to uncontrolled conditions of pH and are being investigated.-P. J. BRUSH.
