With the discovery of the guanylate cyclase-cGMP system in the 1960s, it was anticipated that this system would serve as an important signal transduction and second-messenger system analogous to the adenylate cyclase-cAMP system. However, data to support this hypothesis have only been forthcoming during the past 5-10 years. This paper reviews some of the studies from this laboratory and others that have led to our present understanding of the synthesis of cGMP and its role in smooth muscle relaxation and more specifically its role to mediate the effects of several classes of vasodilators. The regulation of the synthesis of cGMP and some of its effects in several model systems have recently been reviewed in some detail (for a more detailed discussion, see Murad 1986; Leitman and Murad 1987; Waldman and Murad 1987; and the references therein).
