Passage in mice.-One-day-old mice of Webster's VSBS Rockefeller strain were inoculated intracerebrally with 0.01 ml. of passage material. Suspensions and dilutions were made in serum broth-saline (0.9 per cent NaCl containing 9 per cent of broth and 1 per cent horse serum). As a rule the same inoculum was given to all members of a litter except one which was marked and kept as an uninoculated control. For passage one or more mice were killed with chloroform, usually 4 or 5 days after inoculation; their brains were removed aseptically and ground in a TenBroeck grinder with serum broth-saline, 3 ml. /brain. The suspension, after light spinning, was used to inoculate the next mice in the series. As a rule some mice of each litter were observed for 14 days before being discarded. Titration in rabbits.-Brain suspensions and other fluids for titration were, after centrifugation, tested in falling tenfold dilutions in skins of the closely clipped flanks of male Dutch rabbits; 0-1 ml. of each dilution was injected intradermally. The local myxomatous lesions were measured daily, a final reading being taken as a rule on the fourth day; generalised pocks usually appeared by the fifth day, so that later readings were not possible. Titration in egg8.-Three to five 11-day-old eggs were inoculated on the dropped chorioallantoic membrane, each with 0-05 ml. of undiluted brain suspension. They were examined 3 days later, discrete pocks being counted with a lens; titre was expressed as the average number of pocks per inoculum. For very potent samples, dilution was necessary to produtce membranes on which pocks could be counted.
