MethodsThe experiments were performed upon healthy, young CBL rabbits and cats. For animals dosed by gastric intubation, food was withdrawn for 16 hours before administration of the drug. The animals were lightly anesthetized with urethane given intraperitoneally in a dose which averaged 1 Gm. per kg. body weight. Thev were then arranged for the collection· of respiratory tract fluid after methods in use in this laboratorv.'-" The normal output of respiratory tract fluid was measured hourly for a control period of three hours before administration of the drug. Levo propoxyphene was given by subcutaneous injection and bv gastric intubation. For s~ bcutaneous i~ jection, the drug was injected as the hydrochloride dissolved in 1.00 ml. sterilized distilled water per kg. body weight, except in the instance of the highest dose which required 3.00 ml. per kg. Doses given by subcutaneous injection were 9, 27, 81, 243, and 729 mg. per kg. body weight. For oral administration, levo propoxyphene was given as the 2-naphthalene sulfonate, which was suspended in a 0.25 per cent solution of gum tragacanth in distilled water and administered by gastric intubation in a volume of 10 ml. per kg. body weight. Doses given orally were 2, 10, 50, 250, and 1250 mg. per kg. body weight.
