Because platelets express Prpc on their surface, they may constitute a matrix for the conversion of the normal Prpc by the pathological Prpsc and may be a possible carrier of these altered proteins through the body. On the other hand, the fact that platelets shed normal soluble PrP isoforms is important because amyloid proteins, as in Alzheimer’s disease, are thought to derive from soluble precursors or fragments thereof present in biological fluids, which in the disease state undergo conformational change leading to aggregation and deposition in target tissues.’’Furthermore the presence of the normal, soluble prion protein in platelets may allow important questions regarding the functions, the mechanism of Prpc transformation to PrPs°, and amyloidogenesis in prion diseases to be addressed.
