The historical aspects of lathyrism are briefly reviewed, and the radiologic and pathologic aspects of osteolathyrism in the rat are presented. The biochemical and biophysical evidence supporting the concept that the lathyrogens act by blocking intramolecular cross linking in collagen is presented. Normal and lathyritic collagen are identical in the dry state. There may be an increase in the spacing under conditions of maximal hydration of the lathyritic when compared to the normal collagen. This may indicate that the blocking of intramolecular bonds by the lathyrogen allows wider separation of the chains in the collagen molecule than normal.
