METHODSMongrel dogs of either sex weighing lo-30 kg were anesthetized with cr-chloralose-ethyl carbamate (1: lo), 605 mg/kg, following a 24-h fast. The femoral vein was cannulated for intravenous infusion of anesthetic or secretagogue. A midline laparotomy was performed and the viscera were exposed. A gastric branch of the splenic artery was cannulated retrograde for intra-arterial infusion. The antrum was removed to reduce gastrin efl’ects on acid secretion. A wedge from the greater curvature of the fundus was clamped symmetrically with respect to the gastroepiploic vasculature that branches from the splenic artery (insert, Fig. 1). The fundic segment was cut free, exteriorized, and mounted in a Lucite chamber at an angle for drainage by gravity with the blood supply left intact (11). The exposed gastric mucosa was bathed with 150 mM NaCl (saline) for 1 h or more to reach a steady state. The complete chamber assembly is shown in Fig. 1. The mucosal and serosal voltage electrodes detected the spontaneous or clamped PD across the gastric mucosa via saline columns and agar saline bridges connected to calomel electrodes (Radiometer K40 1). Current was passed through the same portion of mucosa from Ag-AgCl mucosal electrodes located in the mucosal chamber solution to Ag-AgCl serosal electrodes located in the serosal saline column. PD was recorded continuously except for periodic measurement of current on an automatic null balance servopotentiometer (Electronik 112, Honeywell), which was modified to allow of separation of potential detection and current passage and to allow for a 20-fold amplification of the original
