EFFECT OF TRAINING AND EXHAUSTION ON HEXOKINASE ACTIVITY OF SKELETAL MUSCLE

Physiol. 27 (5): 691-695. 1969.-The effects of various dynamic exercise programs and of exercise to exhaustion on hexokinase activity in homogenates of guinea pig skeletal muscle were investigated. Hexokinase activity approximately doubled after 9 days of treadmill running at 1.3 km/hr for 30 min/day. Thereafter, exercise was increased to two 30-min bouts/day and hexokinase decreased significantly. After cessation of training the hexokinase activity decreased exponentially, reaching the control-sedentary level after approximately 12 days. When trained animals were run to exhaustion, hexokinase activity decreased significantly. These responses were observed in both the total and soluble hexokinase. Guinea pigs were also exercised 6 days/week for 5 min/day, 15 min/day, or 30 min every other day. After 4 weeks the hexokinase activity approximately doubled with all three training programs. However, the running times to exhaustion ranged from 40 rt 4 to 77= t 13 min for the 5 and 15 min/day groups, respectively. It is concluded that hexokinase activity is very responsive to acute and chronic exercise but the changes induced do not necessarily correlate with the state of training. exercise; aldolasePREVIOUS INVESTIGATIONS(3, 4) from this laboratory have shown that soluble hexokinase activity of skeletal muscle increases with dynamic exercise in adult male guinea pigs. The increase in activity was observed in both “red” and “white” muscle. After 3 weeks of training the activity had increased 1.8-fold compared to the sedentary control animals. An exhaustive run (80 min) in the trained animals produced a significant decrease in hexokinase activity when compared to the usual 30-min bout of exercise. Soluble hexokinase (105,000 X g supernatant) represents approximately 30% of the total activity of skeletal muscle hexokinase with the homogenizing techniques1 used in our laboratory. Therefore, the present study was conducted to investigate further the effects of exercise and exhaustion on skeletal muscle hexokinase. The experiments were designed to answer the following questions.
