Significance: The authors reported the synthesis of a nucleoside containing a rigid nitroxide spin lab (1) via a convergent synthetic strategy. The phenoxazine functionality of 1 was aimed to specifically bind guanine through hydrogen bonds. EPR spectroscopic studies on this spin-labeled nucleoside, spin-labeled oligomer, and spin-labeled DNA duplex clearly demonstrated the limited mobility of spin label within a DNA duplex, indicating its potential application for DNA structural analysis. In addition, compound 1 was found to be easily reduced into fluorophores 2, which are attractive probes for biophysical studies of DNA.Comment: EPR has been an important technique to investigate the structure and dynamics of biopolymers, but this technique requires the incorporation of spin labels at a specific site of the biopolymers. The report described here demonstrated a cleverly designed nitroxide spin label, which can form a base pare with guanine via hydrogen bonds. This feature enables an accurate study of the structure and conformation of DNA duplex containing this spin label. Furthermore, the facile conversion of this spin label into fluorophores offers an opportunity for additional studies of DNA by fluorescence spectroscopy.
