Our interest in quantitative cytochemistry of mammalian blood started years ago, when we were working at Columbia University under the direction of Drs. AW Pollister and L. Ornstein. We were confronted with a problem which at that time had received attention only from a few~ orkers.~,~**~~ Our aim was to ascertain to what extent errors at the ‘‘first stage”(ie, instrument and method of measurement) influenced the “true” spread of a population of Feulgen-stained cells. We used a Pollister-Moses type of cyto~ hotometer~~ and measured Feulgenstained smears of blood and bone marrow of humans and rodents by means of the two-wavelength method of Ornstein and Patau. 12J3
