Hemagglutinin subunits were isolated from three strains of A2/Asian influenza obtained in 1968 before the occurrence of the Hong Kong influenza pandemic, and from three strains of Hong Kong influenza isolated in different parts of the world in 1968, 1970 and 1971. The hemagglutinin subunits were dissociated by treatment with guanidine hydrochloride and dithiothreitol and the light and heavy polypeptide chains were separated by centrifugation. Each of the isolated polypeptide chains was then digested with trypsin and the tryptic peptides were mapped. The maps showed that the polypeptide chains from the hemagglutinin subunits of the “old” A2viruses isolated in 1968 differed greatly in amino acid sequence from the polypeptide chains of the Hong Kong strains. Striking differences occurred in both light and heavy chains. On the other hand, the maps of the three Hong Kong light chains were similar and the maps of the heavy chains were identical with the exception of one or two peptides only. The maps from the three A2/Asian 1968 strains differed in a number of peptides (both in the light and heavy chains) but none bore any resemblance to the maps of the Hong Kong strains.These results suggest that the hemagglutinin subunits of Hong Kong influenza were not derived by mutation from a preexisting human A2strain: it is thought that a more reasonable explanation for their origin is that they came, probably by genetic recombination, from an animal or avian influenza virus.
