The physiological and pathological effects of dimethyl sulfoxide (DMSO) on the skin and health of adult men was extensively studied. Concentrated solutions of dimethyl sulfoxide, 70% and above, markedly increase the penetration of various dyes, steroids, and antiperspirants. The enhancement of penetration is not dependent on irreversible damage to the horny layer. Skin anesthesia was not achieved by 2% solutions of procaine hydrochloride and lidocaine hydrochloride in 90% dimethyl sulfoxide. Dimethyl sulfoxide possesses potent histamineliberating properties at the site of application. The circulating basophils are decreased after extensive topical use but remain within normal limits. Dimethyl sulfoxide is mildly antifungal and antibacterial. Concentrated solutions reduce the resident microflora by 95%. Dimethyl sulfoxide is a promising vehicle for antifungal agents in the treatment of ringworm infections. When injected, dimethyl sulfoxide reduces the clearance of soluble substances from the skin, chiefly by reducing diffusions through the dermis. The resorption of saline wheals is promoted by 90% dimethyl sulfoxide. Dimethyl sulfoxide (90%) neither hindered nor helped experimental thermal burns, contact dermatitis, and ultraviolet burns. Dimethyl sulfoxide (90%) was not systemically injurious when applied once daily to the trunks of healthy subjects for 6 months. Twice daily applications of 90% dimethyl sulfoxide cause a mild scaling dermatitis in certain subjects. The skin rapidly becomes tolerant to the irritant effects of dimethyl sulfoxide even upon continuous exposure. Occlu-sive exposure causes death of the outer epidermis, followed by rapid regeneration despite continued application.
