Macrophages derived from subcutaneous connective tissue were compared in pure cultures with monocytes from blood by: (1) placing both in media of identical composition; (2) studying both in media of differing composition to determine whether metabolic changes modify cytoplasmic structure. Observations were made on mode of locomotion, structure of kinoplasm, characteristics of colonies, food requirements, and susceptibility to arsenious oxide. It was found that (1) monocytes and macrophages become identical in appearance when they live under identical conditions; (2) the form of certain cytoplasmic structures depends on the composition of culture fluid and on the metabolic state of the cells; (3) certain definite physiological properties of both kinds of cells become apparent when grown in pure cultures; (4) knowledge of these fundamental characteristics explains the behavior of these cells within the organism.
