The effect of age on compensatory and "post-functional hypertrophy" in cross-straited muscle.

MethodsThe experiments were carried out in rats aged one month or two years. In the ex periments, in which compensatory reactions in the muscle were tested, the extensor digitorum longus muscle was removed on one side and at the same time the sciatic nerve of this side was cut at an initial operation. 14 days later the m. extensor dig. of the second side was removed. Under these conditions compensatory hypertrophy takes place in the muscle, the percent increase in total protein nitrogen being 12±3.1% in rats 4-5 months old (Gutmann, 1960). Wet and dry weight and total nitrogen were determined. The muscles were dried at 70-80 C in vacuo to constant weight. Neutral fats were extracted with petroleum ether. Total nitrogen was determined after Kjeldahlization of the dry muscle matter by Conway'smicrodiffusion technique (Conway, 1940). In the experiments in which the metabolic changes following stimulation of the muscle were followed, the tibialis anterior muscle was stimulated directly at a frequency of 300/min for 6 min. Immediately, 2 and 4 hours after stimulation the muscles were excised and the absolute amount of noncollagenous protein nitrogen was determined by the biuret method. The amount of ribonucleic acid was determined by Schneider’s method (viz.¿ dk, Gutmann and Vrbovd, 1957). In another series of animals the M. tib. ant. of one side was stimulated at a frequency of 120/min for 3 min 45 sec and the muscles were removed immediately, 2 and 4 hours following stimulation. The levels of glycogen were determined by a modification of the anthrone method (Klicpera, Drahota and¿ dk, 1957).
