These authors have been examining the antibacterial properties of a series of related compounds, of which decamethylene-bis-4-aminoquinaldinium (Dequadin) proved to be the most promising: several salts of this substance of varying solubility were used in different experiments. It is bacteriostatic in very low concentrations for most Gram-positive bacteria and for mycobacteria and fungi, but less active against Gram-negative organisms: serum does not affect this action. It is also bactericidal. Additive effects were obtained when Dequadin was mixed with various antibiotics or cetrimide. It has an intravenous LD50 of only 1.9 mgm. per kgm., the animals appearing to die of respiratory paralysis. Subcutaneous or oral administration was better tolerated, as were various forms of local application. The lives of mice inoculated intra-peritoneally with streptococci or staphylococci could be saved by injecting the drug by the same route 30 minutes later: this is recognized as being a local effect. Hopes are entertained that Dequadin may prove to be of clinical value as a " local chemotherapeutic agent".L. P. Garrod.
