The Specific MR Probes are the key to carry out the target MR imaging. The size, half-life in the circulation, magnetism and combination with the biological marker specifically are the prerequisite factors in the design of targetable probes. In addition, the preparation process is as simple as possible for facilitating mass production. Herein, we report MR molecular probes synthesized using one-pot method.Material and MethodThe carboxylated dextran-coated nanoparticles were conjugated with anti-AFP or anti-GPC3 antibodies through EDC/NHS-mediated reaction to synthesize the probes. The physical and chemical properties of the probes were determined by TEM and DLS, and the relaxivity was compared to uncombined USPIONs using 1.5T clinical MR scanner. The binding efficiency of the antibodies to nanoparticles was measured by an ultraviolet-visible spectrophotometer. In addition, the probes were incubated with targetable cells in vitro.ResultThe results showed that the core mean diameter of the probe was 5–8 nm and average hydrodynamic diameter was 47 nm with a broad slightly distribution. The coupling efficiency of anti-AFP and anti-GPC3 antibodies was 88.8% and 15.9% respectively. There was no statistically significant about shortening the T1 and T2 values of agar solution among USPIO, anti-AFP-USPIO and antiGPC3-USPIO. These two probes can enter the targetable cells, respectively.ConclusionIt is simple and convenient to synthesize the MR molecular probes using one-pot method. The synthesized probes can be used as a high specific agent for experimental study in vitro and to have potential clinical MR imaging
