Marine fouling organisms result in tremendous economic losses to maritime industries and aquaculture facilities around the world. As a result, the development of efficient and environmentally friendly antifouling agents is an urgent task nowadays. Marine microorganisms have been proven to be a potential source of antifouling natural compounds [1]. In our ongoing research on marine fungi from the South China Sea, we have isolated several new secondary metabolites with promising bioactivity, including antifouling activity [2–4]. Recently, we have chemically investigated a gorgonian-derived Eurotium sp. fungus. Six indole alkaloids (1–6) were isolated from the fungal cultures. Among them, compounds 4–6 possess the unusual spiro-anthronopyranoid-diketopiperazine moiety. Compounds 1–3 showed important antifouling activities. Herein we report the isolation, structure elucidation, and antifouling activities of these compounds. The fungus Eurotium sp. was isolated from the inner part of a fresh gorgonian, Subergorgia suberosa, which was collected from the Xisha Islands coral reef in the South China Sea in December 2009. The strain was deposited at the Key Laboratory of Marine Drugs, Ministry of Education of China, School of Medicine and Pharmacy, Ocean University of China, Qingdao, Peoplecs Republic of China, with the Genebank (NCBI) accession number, HM991283. The fungal strain was cultivated in a rice medium (12 g of natural seasalt, 100 g of rice, 0.6 g of peptone, 100 mL of H
