MATERIALS AND METHODSA total of 98 mature female New Zealand white rabbits were used to determine the minimum effective dose of 17p estradiol benzoate which would terminate pregnancy. The animals were injected intramuscularly for three consecutive days according to the schedule shown in table 1. Laparotomies were performed two to 8 days after the last injection. The number of corpora lutea in each ovary and the distribution, size, and appearance of uterine swellings was noted. The rabbit was opened a week to 10 days after the first laparotomp, whenever it was difficult to determine whether uterine swellings contained living or dead embryos. A total of 20 uterine specimens were fixed in Bouin's solution cleared in dioxane, embedded in paraffi, sectioned at 10 p, and stained with either azan or hematoxylin and eosin. In order to determine the effect of estrogen on the deposition of albumen (mucin) around tuba1 eggs, the following procedures were carried out: three animals were given 5 pg of estradiol immediately post coitzlm and killed 24 hours later, 7 animals were given 5 pg of estradiol at one day after mating and killed at 48 hours, 9 animals were injected with 5pg of estradiol at 24 and 48 hours after mating and killed 72 hours post coitum, 9 animals killed at 96 hours were given 5pg of estradiol at 24, 48, and 72 hours after mating. This dosage schedule was later modified to 5 pg of estradiol immediately post coitunz and 5 pg 48 hours later, for reasons explained in the text. A total of 20 control animals were killed at 24 hour intervals during the first 4 days.
