DNA binds Fe(II) in a manner such that OH formation from H2O2 is enhanced as compared to the amount formed in the absence of DNA. Hydroxyl free radical formation was assayed using the spin-trap DMPO (5,5-dimethyl pyrroline-l-oxide). The maximum .**GRAPHIC**. formation occurs at an added Fe(II) per nucleotide ratio of about 1/70. The .**GRAPHIC**. formed H2O2 by the DNA-Fe(II) complex attacked DNA producing aldehydes as determined by the thiobarbituric acid method. These data implicate DNA strand breaks induced by .**GRAPHIC**. attack.
