Phospholamban (PLN, Kirchberger and Tada, 1976), a pentameric integral protein (Mr≈ 6 kDa) of sarcoplasmic reticulum (SR), is an endogenous inhibitor of the SR Ca2+-ATPase. PLN plays a crucial role in excitation-contraction coupling in the heart. Phosphorylation of PLN catalyzed by cyclic AMP-dependent protein kinase activates the SR Ca2+-ATPase in myocardial cells (Tada and Katz, 1982) resulting in the rapid removal of Ca2+ions from the myoplasm to lower the intracellular Ca2+concentration ([Ca2+]i) during each excitation-contraction cycle. The cDNA of PLN has been cloned from the canine heart (Fujii et al., 1987), and subsequently from both cardiac and slow-twitch skeletal muscle of the rabbit (Fujii et al., 1988).
