Reports have indicated that the sensitization of cells to ionizing radiation, as measured by a reduction in colony formation, is related to the amount of 5-bromodeoxyuridine (BUdR) incorporated into the deoxyribonucleic acid (DNA) (1, 2). Studies are lacking, however, on the correlation between radiation-induced chromosomal aberrations and the amount of BUdR incorporated into the DNA. Because cells in the population will not all have the same amount of BUdR incorporated, it is important to measure the incorporation of BUdR in the same cells in which the radiation damage is scored. By making autoradiographs of cells treated with tritiated BUdR (H3BUdR), it was possible to determine in the metaphase cells both the chromosomal damage and, from the number of grains, the amount of BUdR incorporated.
