The .alpha.-MSH variants from rat and human pituitary and rat brain were analyzed in detail by radioimmunoassay interfacing with various chromatography systems. No authentic .alpha.-MSH was detected in extracts of rat brain regions or human pituitary. In these tissues the only .alpha.-MSH variant detected was characterized as des-acetyl .alpha.-MSH. Conversely, acetylated .alpha.-MSH was the predominant form in rat pituitary. In addition to .alpha.-MSH, extracts from both rat and cow pituitary contained an .alpha.-MSH variant with the characteristics of an O-N-diacetyl derivative.
