Immunohistochemical studies have revealed the presence of PYY-like immunoreactivity (PYY-LI) in endocrine cells of the gastrointestinal mucosa of several species, including man, with the highest concentrations in the distal intestine (Lundberg et al. 1982). In a more recent study, evidence for some sparse PYY-positive fibres in the lower brain stem were obtained (Lundberg et al. 1984). We have now reinvestigated a possible occurrence of PYY-like peptide in central neurons, and our results suggest that there exists a small, but widely distributed PYY system with cell bodies in the rostral medulla oblongata projecting to the lower brain stem and spinal cord. Male albino rats (Sprague-Dawley, pathogen free, b. wt 150-200 g, Anticimex, Stockholm) were used. Some rats received an intraventricular injection of colchicine (120/ig dissolved in 20/il, 0.9% NaCl)(Sigma, St. Louis, Mo., USA) and were killed 24-48 h after the drug administration. All rats were anaesthetized with nembutal (40 mg kg-1, ip) and perfused for 6 min via the ascending aorta with Tyrode's solution followed by 10% formalin containing 0.2% picric acid (Zamboni & DeMartino 1976). The brain and spinal cord were dissected out and immersed in the same fixative for 2 h. After rinsing in o. 1 M phosphate buffer containing 10% sucrose for at least 24 h, the brains and spinal cords were cut on a cryostat (section thickness 14/¿ m) and processed for the indirect immunofluorescence technique (Coons 1958).
