Structural Proximity of Mitochondria to Calcium Release Units in Rat Ventricular Myocardium May Suggest a Role in Ca2+ Sequestration

METHODSLeft and right ventricles of adult (4-6 wk old) Sprague-Dawley rats were fixed in glutaraldehyde and osmium and stained en bloc in uranyl acetate. The following distances were measured: Near (NFD) and far (FFD) feet distances, are the smallest distances between the mitochondrial membrane and the nearest and farthest feet (RyRs) within an adjacent CRU. Center distance (CD) is the smallest mitochondria to center of T tubule distance.(N= 100 measurements from 3 rats).RESULTSMitochondria are densely packed in continuous rows between the myofibrils and CRUs are mostly nestled between the ends of two elongated, longitudinally oriented mitochondria (Fig. 1). Most of the mitochondrial profiles (122 over 194 or 63%) are in close proximity of a CRU and over 90% CRUs are in close proximity to a mitochondrion. The actual contacts between the two organelles are probably more frequent than this number, since many are not in the plane of section (Fig 1, C). Near and far feet distances are an average of 33 and 188 nm (Table 1). The center of the T tubule, approximately equidistant from all the channels in the CRU, is also in close proximity to the mitochondria (138-160 nm).
