Estimated Michaelis constants are of the order of 0.4-1 mM for choline and 10-20 uM for acetyl CoA. In cholinergic neurons, choline is supplied via a high affinity uptake system, while acetylcoenzyme A is generated within the cell via a complex series of reactions involving Krebs cycle intermediates. Excess enzyme is normally present in vivo and the amount of synthesis is increased with cholinergic neuronal a~ iv~(I). The rate controlling mechanisms in vivo are still not understood.
