Bacterial mutants have been isolated, calledgroN, that block phage development by interference with the action of the product of the phageNgene. λtrpphages, which depend on theNproduct for the synthesis of tryptophan enzymes, do not make these enzymes ingroNbacteria. Two type of phage mutants have been isolated that can overcome thegroNblock. One type makes an alteredNproduct, the other contains anN-bypass mutation.ThegroNmutation is closely linked to the rifamycin-resistance locus inEscherichia coli. Purified RNA polymerase from thegroNmutant is less activated by salt and more sensitive to rifamycin than is the polymerase fromgro+. This suggests that thegroNmutation produces a structural change in the bacterial RNA polymerase such that it can no longer interact properly with the phageNproduct.
