In order to examine the relationship between steroid hormones and sex differentiation, the initial appearance and development of the steroid-producing thecal cells (SPCs) and steroid-converting granulosa cells in the ovary were observed ultrastructurally during the process of ovarian differentiation in the amago salmon,Oncorhynchus rhodurus. Initial ovarian differentiation was identified by the transformation of oogonia to meiotic oocytes in the gonads of fish at 20–25 days after hatching. SPCs having the ultrastructural features of SPCs of other fishes were first observed on the periphery of clusters containing meiotic oocytes at the onset of ovarian differentiation. At the same time, granulosa cells were also observed inside the basement lamina of the clusters. Some SPCs and granulosa cells developed at the periphery of auxocytes as the ovary developed further. However, extensive development of organelles in SPCs was not observed as the ovary developed further. These results suggest the possibility of steroid production at the time of ovarian differentiation in the amago salmon.
