Although it is generally assumed that peroxisomes arise from dilated regions of endoplasmic reticulum (for reviews see ref. I), membrane continuities between the two compartments have never been illustrated convincingly. Most of the published electron micrographs have demonstrated specialized attenuated “tails” arising from peroxisomes which display striking morphological similarities with profiles of smooth ER. However, these connections have not been analyzed thoroughly as yet in serial sections. Consequently, it is not known, whether they indeed represent smooth ER channels or thin extended peroxisoma1 segments connecting two quite distant portions of the same organelle. In view of the apparent contradictory conclusions regarding the biogenesis of peroxisomes we investigated series of ultrathin sections (80-100 sections per each series] of the mouse preputial gland incubated in alkaline DAB medium. This modified sebaceous gland, which is involved particularly in the biosynthesis of ether glycerolipids,’ exhibits a massive proliferation of peroxisomes3 as well as a considerable degree of peroxisomal pleomorphism during the holocrine secretory cycle.‘.3
