Identification of mouse Ly6H and its expression in normal tissue

TheLy6genes encode glycosylphosphatidylinositol-anchored cell surface glycoproteins that are expressed by myeloid and lymphoid cells and may have a role in T-cell activation. A mouse expressed sequence tag (EST) database was screened for sequences homologous to the humanLy6Hgene. 58 ESTs were identified, 10 of which contained fragments >20 bp long. These sequences were used to generate a predicted cDNA sequence for mouseLy6H. PCR with primers based on this predicted sequence was used to isolate theLy6HcDNA from a mouse brain cDNA library. The gene encoded a protein of 160 amino acids and included 3 translation initiation sites and an N-linked glycosylation site. The gene had 90% identity and 96% similarity to humanLy6H. Northern blotting was used to analyse expression of the mouse gene; the gene was expressed strongly in brain and weakly in eye and reproductive organs. The nucleotide sequence reported has been deposited with the GenBank database with the accession number AF127091.
