Realization of single-molecule sensitivity is crucial for tip-enhanced spectroscopy (TES) and its applications. Plasmonic enhancement and directional emission of TES, which are related to the improvement of detection sensitivity, have been theoretically investigated. Calculated enhancement factors for Raman and fluorescence spectroscopy are of the order 10 10 and 10 4 respectively. Directional TES emission reveals an optimal collection angle of±25º, although tradeoffs must be made between the emission and laser incident angles to ensure high-sensitivity TES. The proposed method enhances detection sensitivity by considering the contributions of plasmonic enhancement and directional emission of TES, useful for the design of high-sensitivity TES platforms.
